چکیده

Multiwavelength spectroscopy is a rapid analytical technique that can be applied to detect, identify, and quantify microorganisms such as Karenia brevis, the species known for frequent red-tide blooms in Florida’s coastal waters. This research will report on a model-based interpretation of UV–vis spectra of K. brevis. The spectroscopy models are based on light scattering and absorption theories, and the approximation of the frequency-dependant optical properties of the basic constituents of living organisms. Absorption and scattering properties of K. brevis, such as cell size/shape, internal structure, and chemical composition, are shown to predict the spectral features observed in the measured spectra. The parameters for the interpretation model were based upon both reported literature values, and experimental values obtained from live cultures and pigment standards. Measured and mathematically derived spectra were compared to determine the adequacy of the model, contribute new spectral information, and to establish the proposed spectral interpretation approach as a new detection method

A Harmful Algal Bloom of Karenia brevis in the Northeastern Gulf of Mexico as Revealed by MODIS and VIIRS: A Comparison

The most recent Visible Infrared Imager Radiometer Suite (VIIRS) is not equipped with a spectral band to detect solar-stimulated phytoplankton fluorescence. The lack of such a band may affect the ability of VIIRS to detect and quantify harmful algal blooms (HABs) in coastal waters rich in colored dissolved organic matter (CDOM) because of the overlap of CDOM and chlorophyll absorption within th...

Microsatellite DNA markers for population genetic studies in the dinoflagellate Karenia brevis

Nine nuclear-encoded microsatellites from an enriched genomic DNA library of the HAB (harmful algal bloom) dinoflagellate Karenia brevis were isolated and characterized. The microsatellites include five perfect (three dinucleotide and two trinucleotide) and four imperfect (two dinucleotide and two trinucleotide) repeat motifs. Gene (haplotype) diversity ranged from 0.153 to 0.750 among a sample...

Comparative analysis of bacterioplankton assemblages from Karenia brevis bloom and nonbloom water on the west Florida shelf (Gulf of Mexico, USA) using 16S rRNA gene clone libraries.

The brevetoxin-producing dinoflagellate, Karenia brevis, forms nearly annual blooms off the Florida west coast, severely impacting the region's ecology and economy. Bacteria are often cited as either promoting or interfering with the development of algal blooms, and thus a detailed study of the bacterioplankton assemblages associated with K. brevis was undertaken. We developed sixteen 16S rRNA ...

Continuous automated imaging-in-flow cytometry for detection and early warning of Karenia brevis blooms in the Gulf of Mexico.

Monitoring programs for harmful algal blooms (HABs) typically rely on time-consuming manual methods for identification and enumeration of phytoplankton, which make it difficult to obtain results with sufficient temporal resolution for early warning. Continuous automated imaging-in-flow by the Imaging FlowCytobot (IFCB) deployed at Port Aransas, TX has provided early warnings of six HAB events. ...

Extraction and analysis of lipophilic brevetoxins from the red tide dinoflagellate Karenia brevis.

Efficient extraction and accurate analysis of lipophilic brevetoxins (PbTxs), produced by the harmful algal bloom (HAB) species Karenia brevis, are essential when assessing the toxicological potential of this dinoflagellate. One of the most commonly used brevetoxin extraction methodologies employs C18 solid-phase extraction (SPE). In this study, C18 SPEC discs were tested for extraction of spik...
